| La caquèxia és una síndrome associada a diversos estats patològics, com creixement de tumors, infeccions cròniques, SIDA o traumes severs; es caracteritza principalment per una pèrdua important de massa corporal. El nostre grup de recerca s’ha interessat per l’estudi dels mecanismes moleculars involucrats en aquesta síndrome, usant tumors experimentals com a model. Els estudis en aquest camp ens han permès concloure que el factor alfa de la necrosi tumoral, una citoquina produïda bàsicament per macròfags, és responsable de la majoria dels canvis metabòlics associats a la caquèxia del càncer, especialment els que estàn relacionats amb un augment de la movilització de les proteïnes musculars. Addicionalment, hem identificat el sistema proteolític que s’activa en aquestes condicions: la ruta proteolítica depenent de l’ATP i de la ubiqüitina. Actualment, estem estudiant la regulació d’aquest sistema proteolític i altres mecanismes involucrats en la caquèxia del càncer (com l’activació de proteïnes no acoblants o l’apoptosi de cèl-lules del muscle esqueletal), per a poder dissenyar noves estratègies terapèutiques per a revertir aquesta condició patològica. |
